Comparing Neurite Outgrowth Using Random and Aligned Electrospun Scaffolds

چکیده
Introduction: Each year in the United States alone, approximately 11,000 individuals suffer from spinal cord injuries [1]. In order to help these individuals, various materials for neural scaffolds are being explored. These materials must mimic the extra-cellular matrix of the spinal cord to help promote cell differentiation and growth for proper recovery. The extracellular matrix is composed of aligned fibers that provide contact guidance for neurite outgrowth. Thus it is expected that neural cells will grow more effectively on aligned scaffolds as opposed to random ones [2]. The purpose of this experiment is to produce random and aligned electrospun scaffolds of three different materials and examine their potential for supporting neural cell growth.

Neurite outgrowth on nanofiber scaffolds with different orders, structures, and surface properties.
Electrospun nanofibers can be readily assembled into various types of scaffolds for applications in neural tissue engineering. The objective of this study is to examine and understand the unique patterns of neurite outgrowth from primary dorsal root ganglia (DRG) cultured on scaffolds of electrospun nanofibers having different orders, structures, and surface properties. We found that the neurit...
متن کاملBioactive peptide functionalized aligned cyclodextrin nanofibers for neurite outgrowth
Guidance of neurite extension and establishment of neural connectivity hold great importance for neural tissue regeneration and neural conduit implants. Although bioactive-epitope functionalized synthetic or natural polymeric materials have been proposed for the induction of neural regeneration, chemical modifications of these materials for neural differentiation still remain a challenge due to...
متن کاملElectrospun poly(caprolactone)-elastin scaffolds for peripheral nerve regeneration
Peripheral nerve regeneration can be enhanced by chemical and mechanical cues for neurite growth. Aligned and randomly oriented electrospun nanofibers of poly(ε-caprolactone) (PCL) or a blend of PCL and elastin were fabricated to test their potential to provide contact guidance to embryonic chick dorsal root ganglia for peripheral nerve regeneration. Scanning electron microscopy was used to ana...
متن کاملElectrospun biomaterial scaffolds with varied topographies for neuronal differentiation of human-induced pluripotent stem cells.
In this study, we investigated the effect of micro and nanoscale scaffold topography on promoting neuronal differentiation of human induced pluripotent stem cells (iPSCs) and directing the resulting neuronal outgrowth in an organized manner. We used melt electrospinning to fabricate poly (ε-caprolactone) (PCL) scaffolds with loop mesh and biaxial aligned microscale topographies. Biaxial aligned...

Objective(s): In this study, the effect of electrospun fiber orientation on proliferation and differentiation of mesenchymal stem cells (MSCs) was evaluated. Materials and Methods: Aligned and random nanocomposite nanofibrous scaffolds were electrospun from polylactic acid (PLA), poly (vinyl alcohol) (PVA) and calcium carbonate nanoparticles (nCaP). The surface morphology of prepared nanofibrou...
